Anti-malarial therapy is given in the following forms -
(A) Casual prophylaxis 
The preerythrocytic phases(in liver) 
Which is thr cause of malaria infection and clinical attacks ,is the target for the purpose.
Primaquine is the casual prophylaxis for all species of malaria, but has not been used in mass programmes ,because of its toxic effects. 
Proguanil is a casual prophylatic ,primarily  p. F ,but is not used in ,because of its weak activitiy against liver stages of p. V.. 
(B) Suppreive prophylaxis -The schizontocides which supress the erythrocytic phase than attack of malaria fever can be used as prophylatic.. 

Best combination used in treatment of malaria -
(1)For uncomplicated malaria
A-Vivax (also ovale, malariae) malaria 
1-Chloroquine + Primaquine
2-quinine +Doxycyline 
B-Chloroquine sensitive falcipuram malaria
Chloroquine +Primaquine 
C-Chloroquine resistance falcipuram malaria 
Artesunate +Meflaquine

(2)For sever/complicated malaria 
Artemether

Arteether
